Pelagic Protocol

Sampling 24th of August:

Sampling preparations were made in the microbiological laboratory above the museum. The necessary equipment was discussed and collected by the course members and the laboratory responsibles. Sterile glass bottle (0,5liter) combined with a sampling rack for surface water sampling was prepared.

The sampling took place on the pier next to the museum during the morning of the 24th of August. Andrei risked his life and dry clothes by sampling the surface water with the sterile glass bottle.

The filled bottle with the water sample was examined by Prof. Boelter and proved to be good. The sample was brought to the laboratory for further investigation. Petri dishes were prepared with Agar for growth experiments. Three petri dishes for colony forming bacteria were prepared by adding 1 millilitre of sample water and stored for growth at room temperature. Three other petir dishes were also prepared for growth experiments (see below). The direct inspection of the water sample by epifluorescence microscopy was performed with two different stains (Acidineorange and DAPI). Aliquots of the sample were filtered on polycarbonat membrane filters (Pore size 0.2 µm) and stained. Acidineorange showed only few organisms and very low content of detritus, which was not used for, further estimates of bacteria. A reference sample stained with DAPI from Baikalsk showed nicely the organisms in the water, bacteria and some algae.

The necessary sample amounts for the further growth experiments were estimated from the epifluorescence microscopy of stained samples (see above). Three polycarbonat filters were prepared by filtering 0.5, 1.0 and 5.0 ml of sample water. The filters are placed on agarplates and incubated at room temperature until next day for further examination.

25 August

11.00-11.30: Phd Pislegina showed us how to process a chlorophyll samples in the hydrobiological laboratory.

11.30-12.30: Post-graduated student of ISU Glushenok told us about the zooplankton community of Lake Baikal. She determined 9 species of zooplankton in the samples, obtained in the point № 1 day before. There were: Notholca intermedia, Keratella cochlearis, Keratella quadrata, Kellicottia longispina, Filinia terminalis, Synchaeta sp., Daphnia longispina, Cyclops kolensis, Epischura baicalensis.

12.30-13.30: There was an interesting and heating discussion about the thickness of fotic zone in Lake Baikal. Dr. Ismest’eva told, that the thickness of fotic zone is equal to 2,5-3 multiply on transparency of water body, measured by Secchi disc, and transparency in Lake Baikal changed from 5 to 44 m.

15.00-19.30: We processed the microbiological samples, obtained in the point № 1. We processed only the samples from surface and from 250 m depth to compare the number of microorganisms. We used the Dapi method and obtained the following results.

19.30 Unfortunately, the fluorescent lamp of microscope exploded and we ended the work.
